Pengaruh zoledronate pada sel tulang tikus sprague dawley pasca ekstraksi gigi: Penelitian histologis pada preparat hematoksillin eosin
L Latar Belakang: Zoledronate adalah obat golongan bisphosphonate yang efektifdalam pengobatan osteoporosis dan diberikan secara intravena. Obat ini bekerjadengan menghambat resorpsi osteoklas dan menurunkan risiko patah tulang akibatosteoporosis pada pria maupun wanita. Penggunaan obat golongan bisphosphonatepada pasien pasca ekstraksi dapat menimbulkan efek samping berupa nekrotikjaringan tulang yaitu bisphosphonates-related osteonecrosis of the jaws (BRONJ).Tujuan: untuk mengetahui pengaruh zoledronate terhadap jumlah osteoblas,osteosit, dan osteoklas. Metode: Jenis rancangan penelitian yang digunakan adalahobservasional analitik dengan rancangan penelitian cross sectional. Sampelpenelitian berasal dari tulang alveolar tikus Sprague Dawley. Seluruh preparatdikelompokkan berdasarkan perlakuan, kemudian dilakukan perhitungan jumlahsel menggunakan perangkat lunak ImageJ dengan perbesaran preparat 40x. Hasilanalisis disalin ke dalam aplikasi SPSS dan dikelompokkan berdasarkan osteoblas,osteosit, dan osteoklas. Data tersebut dilakukan uji statistika dengan metodeANOVA satu jalan. Hasil: Rata-rata jumlah osteoblas dan osteoklas lebih sedikitpada kelompok tulang yang terpapar zoledronate. Sedangkan, rata-rata jumlahosteosit mengalami peningkatan. Pada pengujian Normalitas dan Homogenitas,osteoblas dan osteosit mendapatkan hasil seluruh data berdistribusi normal danosteoklas tidak berdistribusi normal. Berdasarkan hasil pengujian, tidak adaperbedaan jumlah yang signifikan di antara tulang yang terpapar dan yang tidakterpapar zoledronate. Kesimpulan: Pemberian zoledronate mempengaruhi jumlahosteoblas, osteosit, dan osteoklas pasca ekstraksi gigi.
B Background: Zoledronate is a bisphosphonate drug that is effective in treatingosteoporosis and is given intravenously. This drug works by inhibiting osteoclastresorption and reducing the risk of bone fractures due to osteoporosis in men andwomen. The use of bisphosphonate class drugs in post-extraction patients can causeside effects in the form of necrotic bone tissue, namely bisphosphonates-relatedosteonecrosis of the jaws (BRONJ). Objective: to determine the effect ofzoledronate on the number of osteoblasts, osteocytes, and osteoclasts. Method: Thetype of research design used is observational analytic with a cross sectionalresearch design. The research samples came from the alveolar bone of SpragueDawley rats. All preparations were grouped based on treatment, then cell numberswere counted using ImageJ software with 40x magnification. The analysis resultswere copied into the SPSS application and grouped based on osteoblast, osteocyte,and osteoclast. The data was subjected to statistical testing using the one-wayANOVA method. Results: The average number of osteoblast and osteoclast waslower in the bone group exposed to zoledronate. Meanwhile, the average numberof osteocytes increased. In Normality and Homogeneity testing, osteoblasts andosteocytes obtained results that all data were normally distributed and osteoclastswere not normally distributed. Based on the test results, there is no significantdifference in the number of bones exposed and not exposed to zoledronate.Conclusion: Administration of zoledronate affects the number of osteoblasts,osteocytes, and osteoclasts after tooth extraction.
