B Background: tooth discoloration is a common aesthetic complaint and may affect an individual’s self-confidence. one method widely used by the public to manage extrinsic tooth discoloration is the use of whitening toothpaste. whitening toothpastes generally contain abrasive agents or chemical substances, such as hydrated silica and activated charcoal, which function to remove stains from the tooth surface. however, the use of these materials may affect not only tooth colour changes but also enamel surface roughness. objective: to determine the differences in the effects of toothpastes containing hydrated silica and activated charcoal on tooth color changes and enamel surface roughness. method: this study was an in vitro laboratory experimental study with a control group pretest–posttest design. a total of 36 maxillary permanent incisor teeth were divided into three groups: hydrated silica toothpaste group, activated charcoal toothpaste group, and a negative control group without toothpaste. results: toothbrushing using toothpaste containing hydrated silica and activated charcoal resulted in significant changes in the lightness (l*) and chroma (c*) parameters after treatment (p<0.05); however, when compared between groups, no significant differences were found in tooth color changes based on Δe, Δl, and Δc values (p>0.05). in addition, enamel surface roughness increased significantly in both treatment groups compared with the negative control (p<0.05), with the highest roughness observed in the activated charcoal group (0.58 ± 0.15 µm). conclusion: toothpastes containing hydrated silica and activated charcoal induced changes in the lightness (l*) and chroma (c*) parameters; however, no significant differences in tooth colour changes were observed between groups. furthermore, both materials increased enamel surface roughness, with the greatest abrasive effect exhibited by activated charcoal.
